ABSTRACT

Más de nueve millones de sobrevivientes de cáncer de mama en todo el mundo sufren un deterioro en la calidad de vida atribuible a los síntomas de la menopausia relacionados con el déficit de los estrógenos y con los efectos secundarios de la terapia contra el cáncer. La terapia de reemplazo hormonal (TRH) es muy eficaz para controlar estos síntomas en la población general y en las sobrevivientes de cáncer de mama. Sin embargo, la preocupación de la recurrencia del cáncer de mama como resultado del uso de TRH impide que muchos oncólogos utilicen este enfoque en el tratamiento de los síntomas menopáusicos. La evidencia de ensayos aleatorizados, estudios observacionales y meta-nálisis sobre el impacto del uso de TRH en la recurrencia y supervivencia del cáncer de mama sigue siendo controvertida.


More than nine million breast cancer survivors in everyone suffers a deterioration in the quality of life attributable to the symptoms of menopause related to the deficiency of the estrogens and with the side effects of anti-estrogen therapy Cancer. ABSTRACT

SUMMARY INTRODUCTION Although estrogen therapy is widely used against post-menopausal symptoms, it can present adverse effects, including endometrial cancer. Soy isoflavones are considered a possible alternative to estrogen therapy. However, there are still concerns whether isoflavones exert trophic effects on the uterine cervix. OBJECTIVES To evaluate the histomorphometric and immunohistochemical alterations in the uterine cervix of ovariectomized rats treated with soy isoflavones (Iso). METHODS Fifteen adult Wistar rats were ovariectomized (Ovx) and divided into three groups: Group I (Ovx), administered with vehicle solution; Group II (OVX-Iso), administered with concentrated extract of Iso (150 mg/kg) by gavage; and Group III (OVX-E2), treated with 17β-estradiol (10 µg/kg), subcutaneously. After 30 days of treatments, the uterine cervix was fixed in 10% formaldehyde and processed for paraffin-embedding. Sections were stained with Hematoxylin and eosin for morphological and morphometric studies or subjected to immunohistochemistry for detections of Ki-67 and vascular endothelial growth factor-A (Vegf-A). The data obtained were subjected to statistical analysis (p ≤ 0.05). RESULTS We noted an atrophic uterine cervix in GI, whereas it was more voluminous in GII and even more voluminous in GIII. The thickness of the cervical mucosa was significantly higher in GIII, as compared to GI and GII. The cell proliferation (Ki-67) was significantly elevated in the estradiol and isoflavones treated groups, whereas Vegf-A immunoexpression was significantly higher in GIII, as compared to groups GII and GI. CONCLUSIONS Soy isoflavones cause less trophic and proliferative effects in the uterine cervix of rats as compared to estrogen.


RESUMO INTRODUÇÃO Embora a terapia estrogênica seja amplamente utilizada contra sintomas pós-menopausais, ela pode apresentar efeitos adversos, incluindo câncer de mama e endometrial. Assim, as isoflavonas da soja são consideradas uma alternativa possível à terapia estrogênica. No entanto, ainda há controvérsias se estes compostos exercem efeitos tróficos significativos no colo do útero. OBJETIVOS Avaliar as alterações histomorfométricas e imuno-histoquímicas no colo do útero de ratas ovariectomizadas tratadas com isoflavonas da soja (iso). MÉTODOS Quinze ratas Wistar adultas foram ovariectomizadas bilateralmente (Ovx) e separadas em três grupos: Grupo I (Ovx) - veículo (propilenoglicol); Grupo II (Ovx-Iso) - receberam extrato concentrado de Iso (150 mg/kg) e Grupo III (Ovx-E2) - tratado com 17β-estradiol (10 µg/kg); as soluções foram administradas via gavagem por 30 dias consecutivos. Posteriormente, os colos uterinos foram retirados, fixados em formaldeído a 10% tamponado e processados para inclusão em parafina. Cortes (4 µm) foram coradas com hematoxilina e eosina para estudo morfológico e morfométricos, enquanto outros foram submetidos à imuno-histoquímica para detecção de Ki-67 e do fator de crescimento endotelial vascular-A (Vegf-A). Os dados obtidos foram submetidos à análise estatística (p≤0,05). RESULTADOS Observamos a presença de colo uterino atrófico no GI (Ovx), sendo este mais volumoso no GII (Ovx+Iso) e ainda mais volumoso no GIII (Ovx+E2). A espessura da mucosa cervical foi significativamente maior no GIII (Ovx-E2), em comparação ao GI (Ovx) e ao GII (Ovx-Iso). A proliferação celular (Ki-67) foi significativamente mais elevada nos grupos tratados com estradiol e isoflavonas, enquanto a imunoexpressão de Vegf-A foi significativamente maior no GIII (Ovx-E2), em comparação ao GII (Ovx-Iso) e ao GI (Ovx-E2). CONCLUSÕES As isoflavonas da soja causam menos efeitos tróficos e proliferativos no colo do útero de ratas em comparação ao estrogênio.

ABSTRACT

The chemical composition of Mangifera indica L. cv. "Kent" leaves was determined by HPLC-ESI-QTOF-MS/MS. Polyphenolic compounds characterized as benzophenone derivatives were the main components found in extracts (1, maclurin 3-C-(2-O-galloyl)-D- glucoside isomer; 2, maclurin 3-C---D-glucoside; 3, iriflophenone 3-C---D-glucoside; 5, maclurin 3-C-(2,3-di-O-galloyl)---D-glucoside; 6, iriflophenone 3-C-(2-O-galloyl)---D-glucoside; 7, methyl-iriflophenone 3-C-(2,6-di-O-galloyl)---D-glucoside) and xanthones (4, mangiferin and 8, 6-O-galloyl-mangiferin). The estrogenic and antioxidant effects of aqueous extracts from Mangifera indica L. cv. "Kent" leaves on ovariectomized rats were determined by uterotrophic assay and malondialdehyde (MDA) levels in erythrocytes, bone, liver, and stomach. We conclude that the polyphenolic compounds from extracts act as exogenous antioxidant agents against oxidative damage in ovariectomized rats.


La composición química de las hojas de Mangifera indica L. cv. "Kent" se determinó por HPLC-ESI-QTOF-MS/MS. Compuestos polifenólicos caracterizados como derivados de benzofenona fueron los componentes principales encontrados en los extractos (1, isómero de la maclurina 3-C-(2-O-galoyil)-D-glucósido; 2, maclurina 3-C-ß-D-glucósido; 3, iriflofenona 3-C-ß-D-glucósido; 5, maclurina 3-C-(2,3-di-O-galloíl)-ß-D-glucósido; 6, iriflofenona 3-C-(2-O-galloil)-ß-D-glucósido; 7, metil-iriflofenona 3-C-(2,6-di-O- galloyl)-ß-D-glucósido) y xantonas (4, mangiferina y 8, 6-O-galoyil-mangiferina). Los efectos estrogénicos y antioxidantes de los extractos acuosos de hojas de Mangifera indica L. cv. "Kent" en ratas ovariectomizadas se determinaron mediante ensayo uterotrófico y la medición de los niveles de malondialdehído (MDA) en eritrocitos, huesos, hígado y estómago. Concluimos que los compuestos polifenólicos de los extractos actúan como agentes antioxidantes exógenos contra el daño oxidativo en ratas ovariectomizadas.

ABSTRACT

Breast cancer is the most common cause of cancer among women in most countries (WHO). Ovarian hormone disorder is thought to be associated with breast tumorigenesis. The present study investigated the effects of estrogen and progesterone administration on cell proliferation and underlying mechanisms in breast cancer MCF-7 cells. It was found that a single administration of estradiol (E2) or progesterone increased MCF-7 cell viability in a dose-dependent manner and promoted cell cycle progression by increasing the percentage of cells in the G2/M phase. A combination of E2 and progesterone led to a stronger effect than single treatment. Moreover, cyclin G1 was up-regulated by E2 and/or progesterone in MCF-7 cells. After knockdown of cyclin G1 in MCF-7 cells using a specific shRNA, estradiol- and progesterone-mediated cell viability and clonogenic ability were significantly limited. Additionally, estradiol- and progesterone-promoted cell accumulation in the G2/M phase was reversed after knockdown of cyclin G1. These data indicated that estrogen and progesterone promoted breast cancer cell proliferation by inducing the expression of cyclin G1. Our data indicated that novel therapeutics against cyclin G1 are promising for the treatment of estrogen- and progesterone-mediated breast cancer progression.

BACKGROUND: Cimicifuga racemosa is one of the herbs used for the treatment of climacteric syndrome, and it has been cited as an alternative therapy to estrogen. Apart from hectic fevers, dyspareunia and so on, dry mouth also increase significantly after menopause. It has not yet been reported whether C. racemosa has any impact on the sublingual gland, which may relate to dry mouth. In an attempt to determine this, we have compared the effects of estrogen and C. racemosa on the sublingual gland of ovariectomized rats. RESULTS: HE staining showed that the acinar cell area had contracted and that the intercellular spaces were broadened in the OVX (ovariectomized rats) group, while treatment with estradiol (E2) and iCR (isopropanolic extract of C. racemosa) improved these lesions. Transmission electron microscopy showed that rough endoplasmic reticulum expansion in mucous and serous acinar epithelial cells and apoptotic cells was more commonly seen in the OVX group than in the SHAM (sham-operated rats) group. Mitochondria and plasma membrane infolding lesions in the striated ducts were also observed. These lesions were alleviated by both treatments. It is of note that, in the OVX + iCR group, the volume of mitochondria in the striated duct was larger than in other groups. Immunohistochemical staining showed that the ratio of caspase-3 positive cells was significantly increased in the acinar cells of the OVX group compared with the SHAM group (p < 0.05); and the MA (mean absorbance) of caspase-3 in the striated ducts also increased (p < 0.05). Estradiol decreased the ratio of caspase-3 positive cells and the MA of caspase-3 in striated ducts significantly (p < 0.05). ICR also reduced the ratio of caspase-3 positive cells and the MA in the striated ducts (p < 0.05), but the reduction of the MA in striated ducts was inferior to that of the OVX + E2 group (p < 0.05). CONCLUSION: Both estradiol and iCR can inhibit subcellular structural damage, and down-regulate the expression of caspase-3 caused by ovariectomy, but their effects were not identical, suggesting that both drugs confer a protective effect on the sublingual gland of ovariectomized rats, but that the specific location and mechanism of action producing these effects were different.

Background: The decline of estrogen levels increases cardiovascular risk in women. Platelets express estrogen receptors and 17β-estradiol- (E2) can produce a protective effect on thrombus formation. The hydroxylation of cholesterol generates several sterols and 27-hydroxycholesterol (27HC) predominates in circulation. Aim: To evaluate the effect of 27HC as an endogenous antagonist of the anti-aggregating properties of E2 in platelets of postmenopausal women. Material and Methods: Platelet function of postmenopausal women was evaluated ex-vivo. Platelets pre-incubated with 27HC in the presence or absence of E2, were stimulated with collagen. Aggregation was evaluated using turbidimetry using a Chrono-log aggregometer. Results: Collagen-stimulated platelet aggregation was significantly inhibited by E2. The inhibitory effect of E2 on collagen-stimulated platelet aggregation was significantly reversed in the presence of 27HC. Conclusions: The suppressive effect of E2 on platelet aggregation is inhibited by 27HC, which could contribute to increase cardiovascular risk in postmenopausal women.

ABSTRACT PURPOSE: To develop a model for studying cerebrovascular disease prevention in elderly women. METHODS: Sixty 18-month-old Sprague Dawley (SD) rats were randomly divided into an estrogen administration group (EA, n=30) and a non-administration group (NA, n=30); thirty 4-month-old SD rats were allocated to a control group. The EA group received estradiol benzoate starting on the 5th day of a 34-day breeding period, and the serum levels of estradiol (E2), estrogen receptor (ER), and malondialdehyde (MDA) were measured. The MCA of each group was then sampled for viscoelastic experiments. RESULTS: The serum levels of E2 and MDA in the EA group showed significant differences compared to those in the control group (p<0.05), while the difference in ER between the EA and control groups was not significant (p>0.05). The decrease in MCA stress at 7,200 s and the increase in strain at 7,200 s in the EA group showed no significant differences compared to the control group (p>0.05). CONCLUSION: Estradiol administration inhibited the formation of lipid peroxidation products and restored middle cerebral arterial viscoelasticity in aged female rats.

ABSTRACT

PURPOSE:To investigate the effects of classical acupuncture (Ac) and electroacupuncture (EAc) on estradiol-induced inflammation and oxidative stress in health rodents. METHODS: Twenty-four eight-week old female rats were treated with estradiol valerate (EV) 4.0 mg i.m. single dose and randomly assigned to four groups (n=6): G1(control), G2 (Ac), G3 (EAc 2 Hz) and G4 (EAc 100 Hz). After 60 days all rats were anesthetized with chloral hydrate 10% (0.1 ml/30 g weight of the animal) and submitted to Ac/EAc for twenty minutes. The procedures were repeated on days three, five, seven and nine of the study. The equivalent of the human right ST-36 (Zusanli) and SP-6 (Sanyinjiao) acupoints were chosen for needling and electrical stimulation. On the 10th day of the experiment, all rats were anesthetized for collection of blood and tissues (ovaries) samples for biochemical analysis and histological examination. RESULTS:Glutathione (GSH) and malonaldehyde (MDA) concentrations increased significantly in all groups (plasma and ovary) while myeloperoxidase (MPO) activity decreased significantly in all groups compared with control group (G1). CONCLUSIONS:Both classical acupuncture and electroacupuncture decrease systemic and local oxidative stress and ovary inflammation in healthy rats exposed to estrogenic stimulation. EAc enhances lipid peroxidation at systemic and local levels in female rats exposed to estrogenic stimulation.

A ação que o estrogênio desempenha sobre o endotélio depende da integridade deste e consequentemente das características clínicas de cada indivíduo. O uso da terapia hormonal da menopausa (THM) em mulheres com baixo risco cardiovascular geralmente resulta em efeitos benéficos, desde que iniciado em um período próximo da menopausa. Em contrapartida, o seu uso em mulheres com alto risco cardiovascular, como diabéticas ou portadoras de lesões ateroscleróticas já estabelecidas, e ainda naquelas com início da THM em um período superior a dez anos da menopausa geralmente resulta em efeitos maléficos. Nosso objetivo é avaliar os efeitos do estrogênio sobre a função endotelial em mulheres com sobrepeso ou obesidade, ou seja, indivíduos com risco cardiovascular intermediário. Para isso, 44 mulheres na pós-menopausa com idade entre 47 a 55 anos e índice de massa corporal (IMC) de 27,5 a 34,9kg/m², foram randomizadas nos grupos placebo (P) e estrogênio transdérmico (ET). A intervenção consistiu no uso transdérmico de estradiol, 1mg por dia, por um período de três meses. As participantes realizaram avaliação da reatividade endotelial em repouso e após isquemia [pletismografia por oclusão venosa (POV), com medidas do fluxo sanguíneo do antebraço (FSA) e videocapilaroscopia dinâmica do leito periungueal (VCLP), com medidas da velocidade de deslocamento das hemácias (VDH)], dosagens de moléculas de adesão [E-selectina, molécula de adesão intercelular (ICAM-1) e molécula de adesão vascular (VCAM-1)], aferição da sensibilidade insulínica [através do homeostatic model assessment of insulin resistance (HOMA-IR) e área sob a curva (AUC) da insulina durante o teste oral de tolerância à glicose (TOTG)] e mensurações das viscosidades sanguínea e plasmática. As participantes apresentaram idade de 51,77 ± 2,3 anos, IMC de 31,52 ± 2,54 kg/m² e tempo de menopausa de 3 [2-5] anos. O grupo P não apresentou nenhuma mudança significativa em qualquer variável. The action that estrogen plays on the endothelium depends on its integrity and consequently on the clinical characteristics of each individual. The use of menopausal hormone therapy (MHT) in women with low cardiovascular risk usually results in beneficial effects, since it is started in a period close to menopause. In contrast, its use often results in harmful effects in women at high cardiovascular risk, such as diabetic ones or those with established atherosclerotic lesions, and even in those that the beginning of MHT exceeds ten years from menopause. Our goal is to evaluate the effects of estrogen on endothelial function in overweight women, ie, individuals at intermediate cardiovascular risk. For this purpose, 44 postmenopausal women, aged 47-55 years with body mass index (BMI) from 27.5 to 34.9 kg / m², were randomized into placebo (P) and transdermal estrogen (TE) groups. The intervention consisted of using transdermal estradiol 1mg per day for a period of three months. Participants underwent endothelial reactivity assessment at rest and after ischemia [by venous occlusion plethysmography (VOP), with assessment of forearm blood flow (FBF) and dynamic nailfold videocapillaroscopy (DNV), with assessment of red blood cell velocity (RBCV)], measurements of soluble adhesion molecules [E-selectin, intercellular adhesion molecule (ICAM-1) and vascular cell adhesion molecule (VCAM-1)], measurement of insulin sensitivity [by homeostatic model assessment of insulin resistance (HOMA-IR) and area under the curve (AUC) of insulin during the oral glucose tolerance test (OGTT)] and measurements of blood and plasma viscosities. The participants aged 51.77 ± 2.3 years, BMI 31.52 ± 2.54 kg/m² and had a time since menopause of 3 [2-5] years. P group showed no significant change in any variable. ABSTRACT

OBJETIVOS: Avaliar a morfologia dos cardiomiócitos e quantificar o colágeno presente no miocárdio de ratas tratadas com extrato concentrado de soja ou 17β-estradiol (E2). MÉTODOS: Vinte e oito ratas foram divididas em quatro grupos: GCtrl - fase de estro; GOvx - ovariectomizadas (Ovx); GIso - Ovx tratadas com extrato de soja (150 mg/kg, por dia); GE2 - Ovx tratadas com E2 (10 µg/kg, por dia). As drogas e o veículo (0,2 mL de propilenoglicol) foram administrados após 30 dias da realização da ovariectomia, por 30 dias consecutivos. No último dia os animais foram anestesiados, o coração retirado, mergulhado em formaldeído a 10%, e fragmentos dos ventrículos submetidos a processamento histológico, sendo os cortes corados pela hematoxilina e eosina ou pelo picrosirius-red. As análises histomorfométricas (contagem, volume nuclear e quantificação do colágeno) foram realizadas em microscópio de luz e software AxioVision Rel. 4.2, sendo o colágeno determinado pelo programa Imagelab 2000. Os dados foram submetidos ao teste de ANOVA complementado pelo teste de Tukey (p<0,05). RESULTADOS: Notamos maior quantidade de núcleos de cardiomiócitos nos animais dos grupos Ovx e Iso do que no GE2 e GCtrl (GOvx=121,7±20,2=GIso=92,8±15,4>GE2=70,5±14,8=GCtrl=66,3±9,6; p<0,05), sendo o volume nuclear maior nos animais do grupo Ctrl e E2 (GE2=35,7±4,8=GCtrl=29,9±3,6>GIso=26,5±4,5=GOvx=22,4±2,9; p<0,05). Com relação ao colágeno notamos maior concentração no grupo Ovx (GOvx=5,4±0,1>GCtrl=4,0±0,1=GIso=4,4±0,08=GE2=4,3±0,5; p<0,05). CONCLUSÕES: Os estrogênios previnem a diminuição do volume nuclear dos cardiomiócitos e a deposição de colágeno entre as fibras musculares cardíacas. Já a administração de isoflavonas previne somente a deposição de colágeno, o que pode preservar as propriedades mecânicas das fibras cardíacas.


PURPOSES: To evaluate the histomorphometry of cardiomyocytes and collagen present in the myocardium of rats treated with a concentrated extract of soy or 17β-estradiol (E2). METHODS: Twenty-eight rats were divided into four groups: GCtrl - estrus phase; GOvx - ovariectomized (Ovx) and receiving vehicle; GIso - Ovx and treated with soy extract (150 mg/kg per day); GE2 - Ovx and treated with E2 (10 µg/kg per day). The drugs and vehicle (0.2 mL propylene glycol) were administered for 30 consecutive days after ovariectomy. On the last day the animals were anesthetized, the hearts removed, submerged in 10% formaldehyde and fragments of the ventricles underwent histological procedures, and the sections were stained with hematoxylin and eosin or picrosirius-red. Histomorphometric analysis (number and volume of nuclei and quantification of collagen) was performed under a light microscope with AxioVision Rel. 4.2 software, and collagen fibers were quantified using IMAGELAB-2000 software. Data were submitted to ANOVA followed by the Tukey test (p<0.05). RESULTS: We observed a higher number of cardiomyocyte nuclei in animals of the Ovx and Iso groups than in GE2 and GCtrl animals (GOvx=121.7±20.2=GIso=92.8±15.4>GE2=70.5±14,8=GCtrl=66.3±9.6; p <0.05), while the nuclear volume was greater in the Ctrl and E2 groups (GE2=35.7±4.8 GCtrl=29.9±3.6=>GIso=26.5±4.5=GOvx=22.4±2.9; p <0.05). Collagen concentration was higher in the Ovx group (GOvx=5.4±0.1>GCtrl=4.0±0.1=GIso=4.4±0.08=GE2=4.3±0.5; p <0.05). CONCLUSIONS: Estrogen may prevent the reduction of the nuclear volume of cardiomyocytes and collagen deposition between heart muscle fibers, while the administration of isoflavones only prevents the deposition of collagen, which can preserve the mechanical properties of cardiac fibers.

ABSTRACT

BACKGROUND: Estrogens act on estrogen receptors distributed in articular cartilages, synovial membrane, and ligaments, which are thought to be related with degenerative changes. Meanwhile, progesterone is known to have a weak anabolic action on bone formation This study evaluates the effects of estrogen and progesterone hormone on bone/cartilage turnover in ovariectomized (OVX) rats. METHODS: Thirty-five 7-month-old female Sprague-Dawley rats were randomly divided into 5 groups and then ovariectomized bilaterally except the sham control group. The first and the second group acting as controls did not receive hormonal therapy, the third group received estrogen, the fourth group received progesterone, and the fifth group received combination of both hormones 10 weeks after surgery. Evaluations were done using the serum levels of cartilage oligomeric matrix protein (COMP) for cartilage turnover, collagen type I C-telopeptide (CTX-1) and osteocalcin (OC) for bone turnover at 11, 15, 19 weeks after OVX and histology using the Osteoarthritis Research Society International (OARSI) osteoarthritis (OA) cartilage histopathology assessment system. RESULTS: Significantly less cartilage degradation (decreased levels of COMP) was found in the combined hormone treated group in comparison with OVX group. Similarly, both hormonal treatment resulted in increased bone formation and decreased bone resorption i.e., a low overall bone turnover status (decrease in the serum OC and CTX-1 levels). CONCLUSIONS: Combined estrogen and progesterone therapy was found to be convincing in terms of reducing the severity of OA in this experimental model.

ABSTRACT

Background & objectives: The mature fruits of Solanum nigrum contains steroidal glycosides. These are often used as vegetable and there are evidences on tribal use of these fruits as an oral contraceptive. The present study was carried out to evaluate the estrogenic potential of S. nigrum fruits by in vitro and in vivo assays. Methods: Defatted methanol extract of dried S. nigrum fruits was column fractionated and the glycoside positive fractions pooled. Definite concentrations of the fraction were used for in vitro and in vivo assays. The effect on cell viability was analyzed in MCF-7 cell lines by MTT assay followed by in vitro evaluation of estrogenicity by hydroxy apatite (HAP) binding assay. The results were further evaluated in vivo by performing uterotrophic assay in ovariectomized mouse models. Results: At low concentration (40 μg/ml), SNGF induced a dose-dependent increase in MCF-7 cell proliferation, while higher extract concentrations (80-320 μg/ml) caused progressive cell growth inhibition. The competitive binding assay using 3H-E2 suggests that this effect is mediated by estrogen receptor. Mouse uterotrophic assay revealed a classical uterotrophic response in ovariectomized mice in response to S. nigrum glycoside fraction (SNGF). SNGF at a dose of 100 mg/kg of body wt induced the maximum height of luminal epithelial cells which indicated an increase of 30.8 per cent over control (P<0.01) with a correlated increase in uterine wet wt (150% increase over control). Higher doses (250 and 500 mg/kg body wt) of SNGF did not induce any uterotrophic effect. Interpretation & conclusions: Our preliminary data demonstrate the hormone like activity of Solanum glycosides both in vitro and in vivo in mouse, which needs to be further explored to evaluate the possible mechanism and clinical implications.

ABSTRACT

Several plants are used in herbal medicine for family planning. Carpolobia lutea is a medicinal plant in South Eastern Nigeria used for family planning. The study, was designed to investigate the contraceptive, estrogenic and antiestrogenic potentials of the methanolic root extract of Carpolobia lutea in both rats and mice. The contraceptive effect of extract [7 - 21mg/kg] administered by intraperitoneal route for four days in divided doses was tested in mice and rats. Sexually-active males were introduced on day 5 at the ratio of 3F:1M and kept with these females till the end of the experiment. Investigations on the estrogenic and antiestrogenic property of the extract [7-21mg/kg] were done in immature rats that had undergone surgical removal of both ovaries. The effects of the extract [vaginal opening, vaginal cornification, uterine wet weight] were compared with 17-beta-estradiol [0.l micro g/rat/day] as standard drug. Twenty-four hours later, the animals were sacrificed following the last dose and the weights of uterus, kidney, liver and small intestine were recorded. The extract prevented conception in both mice and rats for two gestational periods. Significant changes [p<0.05-0.001] were observed in the length and weight of pups relative to control. There were no abnormalities observed in the pups over thirty days. In ovariectomized immature young rats, the extract showed estrogenic effect [vaginal opening, vaginal cornification and increased uterine wet weight] in low doses while in high doses, it showed antiestrogenic effect. These findings agree with the traditional use of Carpolobia lutea in the control of fertility. The contraceptive property of the extract may be associated with the direct effects of its chemical constituents

ABSTRACT

INTRODUCTION: The role of ovarian hormones and nitric oxide in learning and memory has been widely investigated. OBJECTIVE: The present study was carried out to evaluate the effect of the nitric oxide synthase (NOS) inhibitor, N (G)-nitro-L-arginine methyl ester (L-NAME), on the ability of estradiol to improve learning in OVX rats using the Morris water maze. METHODS: Forty rats were divided into five groups: (1) ovariectomized (OVX), (2) ovariectomized-estradiol (OVX-Est), (3) ovariectomized-L-NAME 10 (OVX-LN 10), (4) ovariectomized-L-NAME 50 (OVX-LN 50) and (5) ovariectomized-estradiol-L-NAME 50 (OVX-Est-LN 50). The animals in the OVX-Est group were treated with a weekly injection of estradiol valerate (2 mg/kg; i.m.). The OVX-LN 10 and OVX-LN 50 groups were treated with daily injections of 10 and 50 mg/kg L-NAME (i.p.), respectively. The animals in the OVX-Est-LN 50 group received a weekly injection of estradiol valerate and a daily injection of 50 mg/kg L-NAME. After 8 weeks, all animals were tested in the Morris water maze. RESULTS: The animals in the OVX-Est group had a significantly lower latency in the maze than the OVX group (p<0.001). There was no significant difference in latency between the OVX-LN 10 and OVX-LN 50 groups in comparison with the OVX group. The latency in the OVX-Est-LN 50 group was significantly higher than that in the OVX-Est group (p<0.001). CONCLUSION: These results show that L-NAME treatment attenuated estradiol-mediated enhancement of spatial learning and memory in OVX rats, but it had no significant effect in OVX rats without estrogen, suggesting an interaction of nitric oxide and estradiol in these specific brain functions.

ABSTRACT

The low incidence of cardiovascular diseases, including hypertension, in premenopausal women has led to the conclusion that ovarian hormones may have a protective effect on the cardiovascular system. We evaluated the effects of ovariectomy and/or estradiol on sympathovagal balance and heart rate variability (HRV) in female spontaneously hypertensive rats (SHR) with tachycardia and compared them to Wistar rats (12 weeks old; N = 8-12). Ovariectomy (OVX) and/or estradiol (10 µg/kg) did not affect basal arterial pressure in either rat strain, but estradiol increased basal heart rate (HR) in OVX SHR (454 ± 18 vs 377 ± 9 bpm). HR changes elicited by methylatropine and propranolol were used to evaluate the sympathovagal balance. Ovariectomy did not affect the cardiac sympathovagal balance of any group, while estradiol increased sympathetic tone in OVX SHR (120 ± 8 vs 56 ± 10 bpm) and sham-operated Wistar rats (57 ± 7 vs 28 ± 4 bpm), and decreased the parasympathetic tone only in OVX SHR (26 ± 7 vs 37 ± 5 bpm). HRV was studied in the frequency domain (Fast Fourier Transformation). Spectra of HR series were examined at low frequency (LF: 0.2-0.75 Hz) and high frequency (HF: 0.75-3 Hz) bands. The power of LF, as well as the LF/HF ratio, was not affected by ovariectomy, but estradiol increased both LF (29 ± 4 vs 18 ± 3 nu in Wistar sham-operated, 26 ± 5 vs 15 ± 3 nu in Wistar OVX, 50 ± 3 vs 38 ± 4 nu in SHR sham-operated, and 51 ± 3 vs 42 ± 3 nu in SHR OVX) and LF/HF (0.48 ± 0.08 vs 0.23 ± 0.03 nu in Wistar sham-operated, 0.41 ± 0.14 vs 0.19 ± 0.05 nu in Wistar OVX, 0.98 ± 0.11 vs 0.63 ± 0.11 nu in SHR sham-operated, and 1.10 ± 0.11 vs 0.78 ± 0.1 nu in SHR OVX). Thus, we suggest that ovariectomy did not affect the cardiac sympathovagal balance of SHR or Wistar rats, while estradiol increased the sympathetic modulation of HR.

ABSTRACT

Estradiol participates in the control of energy homeostasis, as demonstrated by an increase in food intake and in body weight gain after ovariectomy in rats. In the present study, female Wistar rats (200-230 g, N = 5-15 per group), with free access to chow, were individually housed in metabolic cages. We investigated food intake, body weight, plasma leptin levels, measured by specific radioimmunoassay, and the hypothalamic mRNA expression of orexigenic and anorexigenic neuropeptides, determined by real-time PCR, in ovariectomized rats with (OVX+E) and without (OVX) estradiol cypionate treatment (10 µg/kg body weight, sc, for 8 days). Hormonal and mRNA expression were determined at pre-feeding and 4 h after food intake. OVX+E rats showed lower food intake, less body weight gain and lower plasma leptin levels. In the OVX+E group, we also observed a reduction of neuropeptide Y (NPY), agouti-related protein (AgRP) and cocaine- and amphetamine-regulated transcript (CART) mRNA expression in the arcuate nucleus and a decrease in orexin A in the lateral hypothalamic area (LHA). There was an increase in leptin receptor (LepRb), melanocortin-4 receptor (MC4-R), CART, and mainly corticotropin-releasing hormone (CRH) mRNA in the paraventricular nucleus and LepRb and CART mRNA in the LHA. These data show that hypophagia induced by estradiol treatment is associated with reduced hypothalamic expression of orexigenic peptides such as NPY, AgRP and orexin A, and increased expression of the anorexigenic mediators MC4-R, LepRb and CRH. In conclusion, estradiol decreases food intake, and this effect seems to be mediated by peripheral factors such as leptin and the differential mRNA expression of neuropeptides in the hypothalamus.

ABSTRACT

PURPOSE: To recognize the regenerative capacity influenced by the administrating of estradiol. METHODS: 42 female Wistar rats were used, divided into two groups, the control and the experiment group. A resection of approximately 70 percent of the liver was made in the liver of these animals. The control group received an intramuscular injection of one ml of peanut oil. The experiment group were given estradiol hexahydrobenzoate (50µg) diluted in one ml of peanut oil. Calibrations were done after 36 hours and 7 days, using three methods: the formula of Kwon et al.21, to recognize gain in volume, counting of the mitosis figures in five fields and the percentage of positive PCNA nuclei. RESULTS: Gain in volume (mass) was similar in both groups after 36 hours (p=0.1873) and higher in the experiment groups after seven days (p=0.0447). Microscopy showed a similar number of mitosis figures after 36 hours (p=0.3528) and a tendency to be higher in the experiment group after 7 days (p=0.0883). The average of positive PCNA nuclei was higher in the experiment group both after 36 hours (p=0.0009) and 7 days (p=0.0000). CONCLUSION: The estradiol hexahydrobenzoate improved liver regeneration in rats submitted to a 70 percent hepatectomy.


OBJETIVO: Reconhecer a capacidade regenerativa influenciada pela administração de estradiol. MÉTODOS: Utilizaram-se 42 ratos Wistar, fêmeas, divididos em dois grupos controle e experimento. Realizou-se a ressecção de, aproximadamente, 70 por cento do fígado destes animais. Ratos do grupo controle receberam injeção intramuscular de um mililitro de óleo de amendoim, enquanto que os do grupo experimento receberam hexaidrobenzoato de estradiol (50µg) diluídos em um mililitro de óleo de amendoim. Fizeram-se as aferições com 36 horas e 7 dias, com 3 métodos: Fórmula de Kwon et al.21 para reconhecer ganho de volume, contagem das figuras de mitose existentes em 5 campos e percentual dos núcleos PCNA positivos em 5 campos. RESULTADOS: O ganho de volume (massa) foi semelhante nos dois grupos com 36 horas (p=0,1873) e maior no grupo experimento com 7 dias (p=0,0447). À microscopia observou-se número de figuras de mitose em número semelhante com 36 horas (p=0,3528) e tendência a ser maior no grupo experimento com 7 dias (p=0,0883). A média de núcleos PCNA positivos foi maior no grupo experimento tanto com 36 horas (p=0,0009) quanto com 7 dias (p=0,0000). CONCLUSÃO: O hexaidrobenzoato de estradiol favoreceu a regeneração hepática em ratos submetidos à hepatectomia 70 por cento.

ABSTRACT

It has been reported that apical glycocalyx expression of uterine affects embryo implantation. The aim of this study was to determine endometrial glycocalyx and endometrial thickness after estrogen and progesterone injection in hyperstimulated mice at luteal phase. Adult male and female mice were used for induction of pesudopregnancy. The mice were divided into two groups experimental and control groups. Female mice in the experimental group were superovulated and were then mated with vasectomised mice to induce psudopregnancy Experimental group based on hormone injection was subdivided into five groups 1] Estrogen, 2] Progesterone, 3] Estrogen + Progesterone, 4] Antiprogesterone + Estrogen, 5] Sham. Pesudopregnancy in the control group was achieved without any hyperstimulation. The uterines of all groups were collected after 4.5 days of pregnancy and prepared for the assessment of endometrial thickness and endometrial glycocalyx expression. The results showed that estrogen and progesterone injection increased the intensity of PAS reaction, whereas progesterone decreased this. Also our results showed that the endometrial thickness in the sham group was the highest and in the progesterone group was the lowest Our results showed that addition of estrogen to progesterone, compared to P supplementation alone, provided appropriate endometrial conditions to embryo implantation. Hence combination of estrogen and progesterone injection as luteal support hormones can be used in IVF protocols

ABSTRACT

Many studies have shown uncontrolled brain edema to be the cause of disabilities and deaths following head trauma. Current data also suggests that a single administration of estrogen or progesterone can have neuroprotective effects on brain injury. In this study we investigated the combined effect of estrogen and progesterone on brain edema and neurological outcomes following traumatic brain injury [TBI] in female rats This interventional-experimental study was performed on 8 groups of female rats as follows: 1- control, 2-Sham, 3-Ovarectomized trauma [TBI+OVX] 4-Vehicle 5-Physiologic dose of estrogen + physiologic dose of progesterone [E1+P1], 6- physiologic dose of estrogen+pharmacologic dose of progesterone [E1+P2] 7-Pharmacologic dose of estrogen+physiologic dose of progesterone [E2+P1] and 8-Pharmacologic dose of estrogen+pharmacologic dose of progesterone [E2+P2]. Hormones were injected i.p, half an hour after diffuse traumatic brain injury through marmarou model to 2 week old ovarectomized rats. Brain edema [via brain water content], blood-brain barrier permeability [via extra vascular evans blue dye] and neurological outcome [via veterinary coma scale] were measured in this animals The results showed significance decreases of 2.68% and 2.88% in water content in group 8 compared to the vehicle group and group 6 respectively and a significant decrease of 2.29% in water content in group 5 compared to group 6. Evans blue level showed significant decreases of 14.7% and 21.1% in groups 6 and 7 compared to the vehicle group. Neurological scores showed a significant increase of 2.5 and 2 in group 5 compared to the vehicle group and group 3, 1 hour after TBI respectively a significant increase was seen in all groups compared to group 3 at 4 and 24 hours after TBI. Scores showed a significant increase of 1.2 in groups 7 and 8 compared to the vehicle group at 24 hours following the TBI. Based on these results, it can be concluded that combined administration of estrogen and progesterone have beneficial effects on both the reduction of brain edema and the neurological outcomes, the improvement depending on what dose of estrogen is administered with progesterone

ABSTRACT

Estrogen deficiency is associated with unfavorable changes in body composition and abdominal fat deposition. The consequences of these changes lead to metabolic abnormalities and differential fat distribution Seventy female Wistar rats [weight: 170.73 +/- 15.82gr mean +/- SD] were divided into 7 groups: Intact [one group],sham [two groups], ovariectomized [two groups], ovariectomized, receiving estradiol valerate[one group] and ovariectomized, receiving sesame oil [one group]. The intact rats were anesthetized and visceral fat was then taken from the abdominal cavity and weighed immediately. Two weeks after operation, one group of ovariectomized rats and one group of sham rats were sacrified and visceral fat was measured. The estradiol receiving ovariectomized group and vehicle group were given equal volumes of 17 beta-estradiol[30 micro g/kg, sc, 5 d/wk] and sesame oil for 8 weeks, respectively. After 8 weeks, all animals were sacrificied and intra- abdominal fat depots were dissected and weighed. Data were analyzed with one-way ANOVA and post-hoc and using paired t test. The differences were considered significant at P < 0.05. After two weeks of surgery, the ovariectomized rats showed insignificant increase in body weight and visceral fat weight, whereas, after eight weeks, body weight increased significantly in ovariectomized rats [P < 0.05]. Estradiol replacement decreased body weight and visceral fat weight, however this decrease was only significant in body weight [P < 0.05]. The results indicate that estrogen deficiency following ovariectomy leads to increase in both body weight and visceral fat, showing that replacement of this hormone could decrease body weight without affecting visceral fat
